    Abstract

Progesterone is a hormone that relates closely to the reproductive physiology of animals. The progesterone levelsin blood, urine and milk havebeen used to determine pregnancy in cows. In order to detecttheappearance ofprogesterone using specific tests, onemust create hybrid cellsthat secrete monoclonal antibody for this hormone, as one of the most important factors. The methodsof immunization, collection of spleen cellsof BALB/c mice, production of hybrid cell lines and ELISA had been applied. Basedon the use of different types of antigens, mice differently responded to selective progesterone antigens with the concentration of 200 µg/mLper mouse. The highest successful hybridization rate was 91.67%. The study created 5 hybrid cell lines (E4, E3, C6, H3, F10)secreting monoclonal antibodies forprogesterone antigen, 3hybrid cell lines(C12, D7, F11)secreting monoclonal antibodies for progesterone BSAantigenand 3hybrid cell lines(G5, H3, A7)secreting monoclonal antibodies for progesterone-3-CMO: BSA antigen.
